Eficits of Glutamate Transmission in the Striatum F Experimental Hemiballism

چکیده
bstract—Hemiballism (HB) is a quite rare disorder, generally econdary to stroke, neoplasms or demyelinating plaques, lassically considered as almost pathognomonic of a lesion in he subthalamic nucleus (STN). This alteration causes involunary movements in the chorea–ballism spectrum. One theory is hat the output nuclei of the basal ganglia are overinhibited in B, while little is known about the physiological state of the triatum, the major input structure of the basal ganglia. In the resent study, we recorded spontaneous and miniature excitaory and inhibitory postsynaptic currents (sEPSCs, mEPSCs, IPSCs, mIPSCs) from projection neurons of the striatum of xperimental HB. We found a selective reduction of striatal EPSC and mEPSC frequency following chemical lesion of the TN of the rat, suggesting that reduced synaptic excitation of he input structure of the basal ganglia represents a physiologcal correlate of HB. © 2006 IBRO. Muscarinic receptors regulate extracellular glutamate levels in the rat striatum: an in vivo microdialysis study.
Regulation of extracellular glutamate levels by muscarinic receptors in the striatum of unanesthetized rats was investigated by microdialysis. Extracellular glutamate levels were elevated by intrastriatal perfusion of L-trans-pyrrolidine-2,4-dicarboxylic acid (L-trans-PDC), a competitive substrate of plasma membrane excitatory amino acid transporters. The nonselective muscarinic agonist, oxotre...

Cholinergic transmission in the striatum functions as a key modulator of dopamine (DA) transmission and synaptic plasticity, both of which are required for reward and motor learning. Acetylcholine (ACh) can elicit striatal DA release through activation of nicotinic ACh receptors (nAChRs) on DA axonal projections. However, it remains controversial how muscarinic ACh receptors (mAChRs) modulate s...
متن کاملCannabinoids decrease corticostriatal synaptic transmission via an effect on glutamate uptake.
Activation of cannabinoid CB1 receptors reduces glutamatergic synaptic transmission in the rodent striatum and is involved in the normal control of motor function by the basal ganglia. Here we investigated CB1 receptor regulation of glutamate release and uptake and synaptic transmission in the rat striatum. We show that CB1 receptor activation reduces both the release and uptake of [3H]glutamat...
متن کاملMetabotropic glutamate receptor 2 inhibits thalamically-driven glutamate and dopamine release in the dorsal striatum
The striatum plays critical roles in action control and cognition, and activity of striatal neurons is driven by glutamatergic input. Inhibition of glutamatergic inputs to projection neurons and interneurons of the striatum by presynaptic G protein-coupled receptors (GPCRs) stands to modulate striatal output and striatum-dependent behaviors. Despite knowledge that a substantial number of glutam...

Regulator of G-protein signaling (RGS) 4 negatively modulates signaling of several Galpha(q)-coupled receptors, including metabotropic glutamate receptor (mGluR) subtype 5 in neuronal and non-neuronal cell lines. In the brain, both RGS4 and mGluR5 receptors are enriched in the striatum, and their functions have been linked to psychostimulant-induced behavior and synaptic plasticity. However, it...
